Fig. 5: Organelles - DAB reveals catalase in peroxisomes

To demonstrate catalase in peroxisomes, Tetrahymena pyriformis strain GL was incubated in an alkaline medium containing 3,3’-diaminobenzidine (DAB) and hydrogen peroxide (H2O2) for 1, 2 and 4 hours. An electron-opaque DAB reaction product is found after 2 (4) and 4 (5) hours in unstained cells but not after 1 hour (3) of incubation. On uranyl acetate and lead citrate stained thin sections the peroxisomes but not mitochondria stain (6-8). Thus the catalase is present only in the peroxisomes. EMs taken on 12/11/74 by R.
