Paramecium caudatum exposed to the lectin Concanavalin A conjugated to FITC after 5% paraformaldehyde or 10% formalin fixation show the number and location of the nuclei in non permeabilized cells. The nuclear envelopes become labeled. a) This species has one fairly large micronucleus and a much larger macronucleus, both enclosed by a nuclear envelope. The micronucleus tends to lie against the macronucleus. b) During anaphase of micronuclear division the micronucleus elongates within the intact nuclear envelope in the anterior end of the cell. c) As the binary division furrow begins to appear a very long division spindle forms. The macronucleus also elongates. d) In late stages of binary fission the daughter micronuclei separate from the division spindle and the two ends of the elongated macronucleus separate and pinch off so that one product of macronuclear division remains in the proter and the other end remains in the opisthe.
