Fig. 19: acid phosphatase is not found in acidosomes

To see if acidosomes contain hydrolases (digestive enzymes) we incubated cells in Gomori’s medium which in the presence of acid phosphatase (AcPase) and lead will form an electron-opaque reaction product. Acidosomes (ac) surrounding a DV-I are free of reaction product even though a nearby DV, presumably a DV-III, contains abundant reaction product (AcPase). A few vesicles assumed to be lysosomes (ly) also contain some reaction product. This experiment demonstrated that acidosomes are not lysosomes and do not contain digestive enzymes at least not AcPase. EM taken on 4/8/83 by R. Allen with Hitachi HU11A TEM. Neg. 5,000X. Bar = 1µm.
